Regulatory systems in essential metabolism of Streptomyces albidoflavus J1074: discovery and biotechnological application

Project description:

Streptomyces albidoflavus J1074 is among the most efficient platforms for discovery of novel natural products via heterologous expression approaches. Although a number of genetic tools have been developed for J1074, strictly inducible expression systems based on essential metabolites are absent. This severely limits the prospects for multiplexed engineering of J1074, and, hence, chances to provide sustainable access to high-value small molecules. On combining the expertise of the Lithuanian (Bioprocess Research Centre, Kaunas University of Technology) and Ukrainian (Department of Genetics and Biotechnology, Ivan Franko National University of Lviv), partners in genetically encoded biosensors and Streptomyces genetics, respectively, it is proposed to uncover novel regulatory systems embedded in primary metabolism of J1074, such as (but not limited to) fatty acids biogenesis and oxidation, organic acids pathways, sulfur cycle and metal homeostasis

Project results:

The discovered systems will be applied to assist and inform engineering of J1074 to produce acyl CoA and amino acid precursors of bioactive small molecules, bioplastic and biofuels.
